GS-441524 is phosphorylated to nucleoside monophosphate (NMP) by cellular kinase in the cell and then converted to an active triphosphate metabolite (NTP structural analogue). In viral RNA synthesis, active NTP structural analogues act as competitors of nucleoside triphosphate and natural nucleoside (ATP, TTP, CTP, GTP) to participate in RNA transcription. When GS-441524 molecule is inserted into the transcription product, transcription will be terminated in advance, thus inhibiting the viral RNA transcription process.

 

 

Application

GS-441524 is a small molecule known as a competitive inhibitor of riboside triphosphate that exhibits strong antiviral activity against many RNA viruses. It can inhibit the replication of several different types of RNA viruses, such as zoonotic severe acute Chemicalbook Respiratory syndrome (SARS) coronavirus, Middle East Respiratory syndrome virus, Ebola virus, Lassa fever virus, Junin virus, and respiratory syncytial virus, with low cytotoxicity in a wide range of cell lines.
