Sugar spheres contain not less than 62.5 percent and not more than 91.5 percent of sucrose (C12H22O11), calculated on the dried basic, the remainder consisting chiefly of starch. [USP 24].
Sugar spheres are critical excipient for controlled or sustained release drug delivery technologies. They are drug free neutral cores which can be coated by suspension or a solution of the active ingredients with a binder. The fluid bed technology (bottom spray or tangent spray rotor systems) is the best choice for pellets coating, because it allows precise and uniform drug quantity layered on the sugar spheres. For the drug delivery system, a second polymeric film layered on the pellets in order to obtain desired drug release profiles.
